Bone-building discovery could transform osteoporosis treatment

A newly discovered receptor switch that boosts bone growth could transform how we treat osteoporosis, by stimulating the body’s own bone-building machinery using a targeted drug and even mechanical force.

Osteoblasts are bone-forming cells. Osteoclasts, on the other hand, dissolve and break down (resorb) old or damaged bone cells, making room for osteoblasts to create new tissue in areas that are growing or in need of repair. Osteoporosis results from an imbalance between bone formation and bone resorption.

A new study led by the University of Leipzig in Germany has identified a critical regulator of bone formation, the G protein-coupled receptor 133 (GPR133), and a way to stimulate it – opening the door to a new strategy to treat or prevent osteoporosis.

Bone growth is, expectedly, most rapid during childhood and adolescence. Then it slows until our late twenties, when we usually reach peak bone mass. After this, bone density tends to plateau for a time before, generally after age 50, bone breakdown starts to exceed bone formation, gradually decreasing bone density as we age.

Implantable heart patch seals holes and regenerates tissue

A team of researchers have developed a 3D-printed, biodegradable heart patch that seals holes in heart tissue and supports tissue regeneration, showing promise as a safer alternative to current surgical materials.

Worldwide, the leading cause of death is heart attack. Those who survive a heart attack face an ongoing issue with the heart’s efficiency, caused by the death of heart cells and the scar tissue that subsequently forms. In severe cases, the heart wall can rupture, requiring emergency surgery.

Over the years, scientists have developed implantable patches to seal and support damaged heart tissue. In a new study led by ETH Zurich in Switzerland, researchers developed and tested a new type of cardiac patch that not only seals defective areas of the heart, but also heals them.

Smart dressings and stem cells: The future of wound healing

From silver nanofibers to smart, sensory-packed dressings, the next generation of wound-healing technology is blurring the line between biology and engineering. A new review has looked at what’s better, the old or the new.

Humans have been treating wounds for as long as we’ve been getting them. As scientific understanding and technology have improved, so, too, have the approaches we’ve developed to aid wound healing and infection control.

Researchers from National Taiwan University (NTU) have led a review of recent research into advanced wound-healing technologies published over the last decade. Drawing data from clinical trials, animal studies, and lab experiments that explored new materials, biological agents, and smart delivery systems, the researchers compared these next-gen approaches to conventional dressing and therapies.

Turning fat cells into bone heals fractures and reverses frailty

Our fat tissue could be used to make our bones regrow, with scientists successfully using adipose cells to repair spinal compression fractures. It could change how breaks are treated and improve bone strength in diseases such as osteoporosis.

Researchers from Osaka Metropolitan University (OMU) have used stem cells taken from fat tissue – adipose-derived stem cells (ADSCs) – and partially developed them to mature into bone cells. Then, these pre-differentiated cells were assembled into spherical clusters known as spheroids, a formation that scientists have found aids tissue repair and regeneration.

The spheroidal ADSCs were then combined with β-tricalcium phosphate – a biocompatible compound commonly used in bone grafts and dental implants – and delivered to an internal target, which in this case was the L4 and L5 vertebrae of rats with osteoporosis spinal fractures. Micro-CT scans, histology and biomechanical testing was performed at four and eight weeks to gauge bone regeneration and injury healing.

First-ever drug to repair DNA and regenerate damaged tissue is here

After two decades in the making, scientists have cracked the code on a drug that can repair DNA, setting the scene for a new class of therapeutics that can fix tissue damage that occurs through heart attack, inflammatory disease and other conditions.

Researchers at Cedars-Sinai made the breakthrough after first developing a technique to isolate progenitor cells from the heart. These cells, much like stem cells, can form new healthy tissue but in a more targeted way. In other words, these cells taken from the heart can help restore function to that organ.

Medical scientist Eduardo Marbán – then at Johns Hopkins and now Cedars-Sinai – found that heart progenitor cells also have a special mechanism where they send out sacs, known as exosomes, which carry molecules of DNA, RNA and protein between cells and can repair and regenerate damaged tissue.

“Exosomes are like envelopes with important information,” said first author Ahmed Ibrahim, PhD, MPH, an associate professor in the Department of Cardiology in the Smidt Heart Institute. “We wanted to take apart these coded messages and figure out which molecules were, themselves, therapeutic.”

FDA approves first dual-action eye drop for age-related vision loss

The US Food and Drug Administration has approved a landmark eye drop that uses a combined dose of medication to restore age-related near-sightedness, without the need for surgery, for a longer duration than anything else on the market – and with less side effects.

Known as Yuvezzi, the eyedrops developed by Tenpoint Therapeutics treat presbyopia, a very common condition that affects us with age, making it more difficult to see text and other things up close. It's now the only FDA-approved drop that combines two active ingredients – carbachol and brimonidine tartrate.

Presbyopia affects more than 100 million Americans and two billion people worldwide, with its onset generally beginning in our 40s. Mostly it's managed with reading glasses, contact lenses or invasive surgery, but until recently there hasn't been any sort of medication to help manage it.
