Vaccine Shows Promise Against Aggressive Brain Cancer

Introduction:

(Futurity) In patients with an especially aggressive form of glioblastoma, the vaccine caused no serious side effects and prolonged patients’ overall survival compared to historical outcomes after standard-of-care surgery and chemo-radiotherapy. One long-term survivor remains recurrence-free nearly five years later.

The results of the phase 1 trial, conducted at Siteman Cancer Center, based at Barnes-Jewish Hospital and Washington University in St. Louis Medicine, appear in Nature Cancer.

The study was led jointly by Mass General Brigham and Geneos Therapeutics, a Philadelphia-based biotechnology company.

“We are extremely encouraged by these results,” says Tanner M. Johanns, lead author of the study and an assistant professor in the Division of Oncology in the medicine department at WashU Medicine.

“This kind of vaccine is a first for glioblastoma, and it is exciting to think how we can leverage this individualized therapeutic DNA cancer vaccine platform to make a positive impact on the lives of patients who are fighting this disease. Additionally, combination therapies leveraging this personalized platform are currently being investigated at WashU to test if outcomes may be improved further.”

CRISPR enzyme precisely detects and shreds DNA in cancer mutations once considered 'undruggable'

In 2020, Jennifer Doudna won the Nobel Prize in chemistry for her work on the CRISPR-Cas9 gene-editing technology that allows scientists to precisely modify DNA by cutting it at specific locations. Six years later, a new study in Nature by a team led by Doudna has uncovered a powerful new approach to selectively kill cancer cells using a CRISPR enzyme called Cas12a2.

Once the enzyme detects cancer-specific genetic signatures, it begins to shred chromatin—a mixture of DNA and proteins that forms chromosomes—within the targeted cell.

Many cancers are driven by mutations in tumor suppressor proteins such as TP53, which is altered in nearly half of all cases. Yet these mutations have remained difficult to treat because they lack binding pockets for traditional drugs to latch onto. As a result, many cancer-causing mutations have long been considered undruggable.

China’s first AI-powered cancer vaccine production line set to launch in Beijing

China has broken ground on what developers say is the country’s first production line for AI-assisted personalised tumour vaccines, raising hopes for millions of new cancer patients every year – a disease that ranks as the nation’s second-leading cause of death.
By October, Beijing-based Likang Life Sciences is expected to complete a new drug research and manufacturing centre in the Beijing Economic and Technological Development Zone, with a total investment of about 110 million yuan (US$16.1 million), according to the district government.

The facility will house cell therapy research laboratories together with a production line of the company’s flagship product, LK101, a personalised cancer vaccine that analyses each patient’s tumour DNA to pinpoint the specific genetic mutations driving the disease. With AI, the company said the procedure could be completed in a day.

Sitting for more than 30 minutes at a time linked to a higher risk of cancer death

Sitting for longer than half an hour at a time each day raises the risk of dying from cancer, a study suggests.

Researchers who tracked more than 90,000 people over a decade found that sitting or lying down while awake for more than 30 minutes in one period each day was associated with an increased risk of cancer death. The risk increases for every additional hour of continuous inactivity, the findings suggest.

However, the researchers also found breaking up periods of sedentary behaviour longer than 30 minutes with bursts of physical activity could help reduce the risk. Getting up every half-hour, even for a short walk around the office, could do wonders for your health, they said.
